It did not work immediately.
The above info is the content of my dsm.conf file to change ports.

Does a machine have to be restarted to make this take effect?

Run the command below.
/etc/rc.d/init.d/dsm restart

What is the command for changing the DSM mail port from 3001?

The webmail client is run directly from Apache. Not under dsm itself, so this would probably not be a good option for a dsm.conf configuration setting.
